Jol admitted Steed Malbranque left an outstretched leg when the United winger burst into the area just before the break at White Hart Lane, but replays showed there was no contact.
Dutchman Jol did not accuse Ronaldo of diving, but did not believe referee Mark Clattenburg should have awarded the spot-kick.
Ronaldo slammed in his penalty and then set up Paul Scholes after Nemanja Vidic doubled the lead after the break.
Ryan Giggs completed the 4-0 win as United responded in style to Chelsea closing the gap following victory on Saturday over Charlton.
They finished comfortably — although John O’Shea was in goal after Edwin van der Sar was taken off with a broken nose — but the turning point was when the scores were level and Ronaldo earned his penalty.
“I don’t think it was a penalty but Steed Malbranque stuck his foot out and pulled it in again,” said Jol.
“He took advantage of that but I don’t know. It didn’t help.
“The referee must make a decision, he is a human being. He was standing there with a good view of it.”
Jol added: “I wouldn’t say Ronaldo was diving. It was a difficult situation.”
Spurs fans were shown replays of the incident at half-time and jeered Ronaldo even louder in the second half. The Portugal winger has dealt with constant boos from fans for his role in Wayne Rooney’s sending off at the World Cup but his form has put United on course for their first title since 2003.
His dazzling work on the flanks continued, with United boss Alex Ferguson adding: “It’s fantastic, the boy has come on great, has the courage to play and always wants the ball.
“At the start of the game I thought he was a bit careless but I think he improved when he got more of the ball. He’s that type of player, you have to service him and get the ball to him and get him working in a game.
“He’s not the type of player who can play on the fringes, he’s got to be involved all the time.”
Rooney took a knock in the second half and Ferguson will assess the striker before deciding whether he will go on England duty this week.
“Wayne was injured in a challenge and he limped around for a bit but I had to take him off,” said Ferguson. “I’ll assess what he’s like in the morning but he may be a doubt for the game on Wednesday.”
With their superior goal difference, United are effectively seven points ahead of champions Chelsea.
Ferguson added: “The scoreline is very good, to come to Tottenham and win 4-0 is an exceptional scoreline.
“In fairness to Tottenham they made a lot of chances in the second half. We maybe were a bit lucky on a couple of occasions. I was a bit concerned and will have to look at a video of that.
“We looked when we went in front we were starting to get careless and casual at the back, you can’t do that. Zeroes against your name is the name of the game. No goals against in the run-in when you’re in the title race is very important to us.”
